Effects of Ocrelizumab on Dental Pulp Derived Mesenchymal Stem Cells

Multiple sclerosis (MS) is a chronic autoimmune central nervous system (CNS) disease characterized by inflammation, demyelination and axonal damage. The most important goal in MS treatment to provide neuroregeneration. However, current treatments only work slow the progression of MS. Ocrelizumab humanized anti-CD20 monoclonal antibody used In this study, it was aimed investigate effects on proliferation differentiation potential dental pulp-derived stem cells (DP-SC). For purpose, three different (1.5 µg/mL, 15 µg/mL 150 µg/mL) doses were added normal culture medium, then analyzes at 24, 48 72 hours performed with WST-1 assay. for 24 observed dose-dependently reduce cells. At hours, efficacy drug lost low doses. Similarly, dose-dependent decrease expression stemness marker, Rex1, detected. adipogenic marker ADFP, osteogenic RUNX2, chondrogenic SOX9, neurogenic TUBB3 evaluated determine influence under effect. While SOX9 expressions increased manner, there ADFP parallel dose. According results ocrelizumab might have support regenerative supporting DP-SCs osteogenic, chodrogenic neurogenic.
